If you follow a big daddy without a little sister around for a while, he'll eventually get a new one from the holes.
I haven't personally seen it happen but I assume the opposite is true too, making it somewhat dynamic.
Big daddies will always spawn, but little sisters will stop spawning once you've rescued/harvested the amount that the level lets you.

If you're purely looking for little sisters to rescue/harvest, pretty much yes.
Probably would be better to travel around the level rather than waiting in one spot. They're pretty noisy, too, easy to locate because of that.
I'm not sure what the criteria is for them spawning with a little sister vs. spawning without one and getting one from a hole. Might be random chance, might be that the first few big daddies you encounter in the level begin with little sisters and the rest are random chance. No idea.
Big daddy spawns are somewhat common though, even if they spawn without a little sister, so shouldn't be too hard.

I think there was only one or two times in which I had to go out of my way to find all the little sisters in a level. Usually the "missions" loop you around enough to encounter enough big daddies without having to seek them out.

Pretty sure the holes aren't actually connected in any way, game-mechanics wise.
So, one hole's little sister status most probably won't affect another's.

You won't find one per hole, no. Too many holes in the level, the harvestable amount would be way more than the level limit.

Don't worry too much about which hole has which little sister, pretty sure it doesn't matter and there is no such system to begin with. Just worry about finding a lonely big daddy and following him. The rest should be smooth sailing.
