There are two different kinds of qui and que.
If you mean the interrogative pronouns, they're straightforward qui = who and ue = what.

Qui est venu avec toi ? = Who came wiht you?
Que lis-tu ? = What are you reading?
However, if you mean the relative pronouns, their uses depend on the grammatical role of the word before the pronoun in regards to the subordinate clause. So, qui marks the subject of said clause while que denotes its object. Unlike the interrogative pronouns, their use does not depend on the animation/inanimation distinction like in English.
La personne qui vient d'arriver est mon frère = The person who just arrived is my brother
La clé qui est tombée sur la route est à mon voisin = The key that dropped on the street is my neighbor's
Le monsieur que tu as rencontré est mon père = The mister whom you met is my father
Le livre que tu m'as apporté est abîmé = The book that you brought me is damaged
