Am I able to get an icon to appear permanently as a desatured icon, then turn to full colour icon when the debuff is applied with 1 trigger?

Yes. Show = Always. Then Conditions to control the Desat

only thing is i can only see the weak aura when i have a target, is there a way to have it on 24/7?

If you're using the "target" unit then there's also a setting to show without a valid unit in the trigger settings
