I have just set up a ender 3 v3 ke. Did full z offset and bed calibration. Still i get some individual lines that are higher than the rest (see image)

What could be the cause and how to solve it?

I recently relocated the spool and am routing through a tube now, so maybe it's an extrusion problem?
I just did 2 smaller prints with this printer, so i have limited "before" experience with this setup.

Over extrusion on first layer. Check flow, diameter, nozzle size, esteps
