My brother and I use separate accounts on separate Xbox’s, I was told by Xbox support I can share a game pass ultimate subscription with him by setting a certain Xbox to the home Xbox. Although he already has an xbox live subscription, if I get the game pass ultimate subscription and share it with him, what will happen to the existing subscription?

If you get game pass ultimate and set your brother's console as your home Xbox, he'll be able to play online and all the games on game pass utilizing your subscription while on his Xbox (as it is your home Xbox).
He will not however get any discounts on full game purchases that are associated with having game pass ultimate, nor will he be able to use the cloud service, as those are tied directly to the account.
His existing subscription will be untouched and continue until it runs out as he isn't actually getting your subscription, just access to the games and the ability to play online through your home Xbox setting.
