Sometimes unlocks seem delayed, like they don't happen until you start a new round/run, but this one remained locked even when starting a new run. Unlocks should happen and give you the notification the instant they occur.
Update: It unlocked after failing another run and going to the main menu. My total sold was still 20.

The counter counts towards challenges and seeded runs, while you need 20 purchased without seeded runs or challenges, so you just need to buy more to unlock it.
