General purpose tip on key setup, applies to both keyboard and controller:
- actions you press a lot show be extremely close to your finger's neutral position and easy to press. For example: your bread and butter combo
- bread and butter combo should flow smoothly when pressed, so try not to put your 3 step combo in weird layout, like q, c, then g
- stuff you rarely press can be parked further away, like your tank stance, or your limit break

I try to think of setting up my hotkeys in tiers. Like tier 0 would be the bread and butter things, like mentioned above, your basic combos and stuff. Tier 1 would be frequently used oGCDs, buffs and other shenanigans and then it just goes from there. The higher the tier, the further away I put the hotkey

you can certainly add target logic to macros, but that's really narrow to call it "logic"
I used to have macros to heal over mouse over target, but I realized I sometimes ended up healing myself due to how macros work (slowly execute line by line) and so I prefer to queue my heals and ended up deleting my macros
